STL Therapy V3.1 introduces a precision cancer-therapy platform built on ultra-stable circularized e-circRNA, integrated with a Rolling Circle Translation (RCT) mechanism that enables perpetual intracellular amplification of therapeutic proteins.
The system activates only when all three layers of an AND Logic Gate are satisfied (Toehold Switches sensing tumor-specific microRNAs and micro-environmental cues), triggering the release of a combined payload consisting of Cas13b RNA-degradation effectors, BiTEs for immune redirection, and anti–PD-L1 shRNA.
The therapeutic cargo is delivered via a hybrid Exosome–LNP vector (functionalized with CD47/iRGD) to maximize stealth and deep-tissue penetration, while incorporating a Degron safety switch that enables immediate shutdown of the therapy through administration of an exogenous oral drug.
